Title: The effect of acupuncture on hippocampal dentate gyrus LTP in vascular dementia model rats in vivo
Abstract: Objective To sdudy the effect of acupuncture on the learning ability, memory and long-term potential(LTP) of hippocampal dentate gyrus in vascular dementia(VD) model rats in vivo. Methods Forty-five rats were divided into control, model and acupuncture groups. The experiental VD animal model was established with improved four vessel occlusion method.Rats in acupuncture group were given acupuncture at ear acupoint kidney and heart daily. The capabilities of learning and memory of the experimental animals were assessed through Morris water maze behavior test.LTP in the hippocampal dentate gyrus was recorded as electrophysiologic index for evaluation of learning and memory. Results Compared with those of model group, the acupuncture group rats′ average escape latency in place navigation test were shorter, the swimming time in third quadrant in spatial probe test longer and times of traversing platform increased(all P0.05).After high frequency stimulation, compared with those of model group, the acupuncture group rats′ the population spike(PS) latency was shorter, and the excitatory postsynaptic potential(EPSP) slope and PS amplitude increased(all P0.05).Conclusions Acupuncture could facilitate the induction and maintenance of LTP in hippocampal dentate gyrus through influencing some synaptic taches in vivo.
