Abstract:Objective To investigate the effect and mechanisms of Glycyrrhizic acid(GA) on human glioma U251 cells.Methods U251 cells were treated with GA at various concentrations,the effects of GA on cell proli...Objective To investigate the effect and mechanisms of Glycyrrhizic acid(GA) on human glioma U251 cells.Methods U251 cells were treated with GA at various concentrations,the effects of GA on cell proliferation and colony formation of U251 cell were measured by CCK8 assay and plate clone-forming test,respectively.And then,cell apoptosis was detected by Hoechst 33258 staining and flow cytometry with Annexin V / PI double staining.The expression of p65 in the nucleus,the active subunit of nuclear factor kappa B(NF-κB),was measured by Western blot and immunofluorescence.Results Compared with the control group,U251 cell survival rate and colony formation were decreased in a time-and dose-dependent manner.The apoptotic ratio of GA group was significantly higher than that of the control group.Besides,the expression level of p65 in the nucleuswas reduced after GA treatment.Conclusion GA may inhibit the proliferation and induce apoptosis on human glioma U251 cells via NF-κB mediated mechanism.Read More
