Abstract:Objective To investigate the effects of glucagon like peptide-2 on intestinal bacteria translocation in rats with sepsis.Methods 60 healthy male Sprague-Dawley rats,were randomly divided into three ex...Objective To investigate the effects of glucagon like peptide-2 on intestinal bacteria translocation in rats with sepsis.Methods 60 healthy male Sprague-Dawley rats,were randomly divided into three experimental groups: normal control group(group A) were injected with normal saline intraperitoneally,LPS group(group B) and LPS+GLP-2 group(group C) were intraperitoneally treated with LPS and LPS plus GLP-2 respectively.All rats were sacrificed at 6 h after injection,terminal ileum of each rat was collected for the observation of the pathologic changes of intestinal mucosa under transmission electron microscope.The expression of occludin was analyzed by immunohistochemistry and western blot.Homogenated tissues of mesenteric lymph nodes,liver and lung were cultured to determine bacterial tranlocation.Results ①Microvilli and tight junctions were intact in group A.Enlargement of tight junctions were observed in group B and microvilli were thin,rare and shed.The rough endoplastic reticulum,mitochondria,and glycogen particles were damaged.The above changes were alleviated in group C.②The content of occludin was obviously lower in group B than that in group A(P0.01).While the content of occludin was significantly increased in group C in comparison with group B(P0.01).③In comparison with group A,bacterial translocation rate in mesenteric lymph nodes,liver,lung in group B was significantly increased(P0.01).As compared with group B,bacterial translocation rate was makedly decreased in group C(P0.01).Conclusion Glucagon like peptide-2 can reduce the degree of intestinal mucosa damage and bacterial translocation with sepsis,it played an important role in protecting intestinal mucosa barrier.Read More
