Title: Functional identification of a vesicular ATP transporter, VNUT, mediating ATP release in human epidermal keratinocytes
Abstract: The skin is one of the largest organs of the body and the first site of exposure to various harmful environmental factors such as infection and UV radiation. In response to cell damage by irritant chemicals or physiological mechanical stimulation, ATP is immediately released from epidermal keratinocytes. However, the mechanisms of its release remain largely unclear. The aim of this study is to determine mechanisms underlying release of ATP, especially focusing on VNUT, a recently identified vesicular nucleotide transporter, in normal human epidermal keratinocytes (NHEKs). When stimulated with ionomycin, NHEKs released ATP, which was inhibited by either BAPTA–AM or bafilomycin A, suggesting the involvement of Ca2+ and vesicular H+–ATPase in the release. The mRNAs and proteins for VNUT were expressed in NHEKs. When VNUT was knocked down by small interfering RNA, the ionomycin–induced ATP release was almost abolished in NHEKs. These results suggest that NHEKs release ATP by a mechanism of exocytosis, for which VNUT has a critical role in forming ATP–enriched vesicles.
