Title: Effects of Polygonum bistorta L.-413 on the Relaxant Mechanism of Isolated Thoracic Aortic Rings in Rats
Abstract: [Objective] To research the effects of Polygonum Bistorla L-413 on the relaxant mechanism of isolated thoracic aortic rings in rats.[Method] Effects of P.Bistorla-413 on the contraction of vascular smooth muscle induced by norepinephrine(NA) were studied under the Ca+ contained and Ca+ free bath solutions.Time-dependent manner of the relaxing effects of P.Bistorla-413 was investigated.Effects of P.Bistorla-413 on the contraction of vascular smooth muscle induced by 40 and 80 mmol/L KCl solutions were researched.[Result] P.Bistorla-413 had relaxing effects on the contraction of vascular smooth muscle induced by NA in a concentration-dependent manner.P.Bistorla-413(100 μmol/L) reached the maximum relaxing effects at 30 min;P.Bistorla-413 in Ca+ free group had greater inhibitory effects on the contraction of vascular smooth muscle induced by NA than that in Ca+ contained group.P.Bistorla-413 had inhibitory effects on the contraction of vascular smooth muscle induced by 40 and 80 mmol/L KCl solutions.And the dose-response curve of two obviously moved up.[Conclusion] P.Bistorla-413 had relaxing effects on the vascular smooth muscle.And the action mechanism might be that P.Bistorla-413 could promote the synthesis and release of NO,open the calcium-activated potassium channels,and inhibit calcium influx and inner calcium release of smooth muscle cell.
