Pretreatment of the animals (24 hours prior to the experiment) with increasing doses of the reserpine-type alkaloids produced, in the heart-lung preparation, a graded reduction of the rate-increasing effect of a challenging dose of 3 mg reserpine. Since reserpine and related alkaloids are well known to deplete the cardiac stores of norepinephrine, the decreased effectiveness of the challenging dose of reserpine may be taken as an indicator of depletion. From the dose-response curves, estimates of the relative potencies were obtained. Desenpidine was about equipotent with reserpine; rescinnamine, raunescine and syrosinigopine had about one-third the potency of reserpine, whereas isoraunescine was much less potent.Read More
