Title: Induction of drug metabolizing enzymes in the liver of diabetic mice
Abstract: Les effets de deux inducteurs classiques, le phénobarbital et le 3-méthylcholanthrène, ont été comparés sur quelques activités de phase I (monooxygénases) et de phase II du métabolisme des xénobiotiques, ainsi que sur le métabolisme du benzo(a)pyrène, chez des souris atteintes de diabète soit génétique (ob/ob), soit chimique (streptozotocine). 1) Chez les souris ob/ob, les activités basales et l'inductibilité des enzymes de phase I et de phase II, ainsi que le profil électrophorétique des protéines microsomiques, ne sont pas notablement différents de ceux de souris de souche sauvage traitées de manière analogue. 2) L'existence possible d'une forme de cytochrome P 450 commune aux microsomes de souris non induites traitées par la streptozotocine, et à ceux de souris non diabétiques traitées par le phénobarbital, pourrait rendre compte de l'augmentation des enzymes de «type phénobarbital chez les animaux atteints de diabète chimique. 3) Cette augmentation des activités monooxygénasiques dues au traitement par la streptozotocine est en partie supprimée par le 3-méthylcholanthrène, probablement en raison d'une dilution des formes de cytochrome P 450 de «type phénobarbital, par le cytochrome P 448 induit par le 3-méthylcholanthrène. 4) La formation accrue des métabolites les plus cancérigènes du benzo(a)pyrène, et la légère diminution d'activité des enzymes de conjugaison, sont susceptibles d'additionner leurs effets délétères chez les animaux atteints de diabète chimique et traités par le méthylcholanthrène. The effects of two classical inducers, phenobarbital and 3-methylcholanthrene, have been tested on some liver microsomal drug-metabolizing enzymes (monooxygenases and phase II enzymes) and on benzo(a)pyrene metabolism in genetically (ob/ob) and chemically (streptozotocin) diabetic mice. 1) In ob/ob mice, the basal activities and the inducibility of phase I and phase II enzymes, as well as the electrophoretic pattern of microsomal proteins, were not notably different from those of similarly treated lean mice. 2) A possibly common form of cytochrome P 450 present both in microsomes from steptozotocin-diabetic non-induced mice and in those from phenobarbital-treated non-diabetic mice could explain the increased «phenobarbital-like enzyme activities in chemically diabetic animals. 3) The increase of monooxygenase activities produced by streptozotocin treatment is partially depressed by 3-methylcholanthrene, probably as a result of the dilution of «phenobarbital-like cytochrome P 450 forms by 3-methylcholanthrene-induced cytochrome P 448. 4) The increased formation of the most carcinogenic metabolites of benzo(a)pyrene, and the slight decrease of phase II conjugation enzyme activities, may add their deleterious effects in 3-methylcholanthrene-induced streptozotocin-diabetic animals.
