Title: Contractility of adult rat/mouse cardiomyocytes expressing human cardiac troponin I_wt and –R145G
Abstract: We investigate the effects of amino acid exchanges in cardiac troponin I (cTnI), known to induce familial hypertrophic cardiomyopathy (FHC) on the contractility of adult cardiomyocytes isolated from rat or mouse left ventricles. The extent, rate and duration of sarcomer shortening and its relaxation rate are analysed in response to β_, β_1 and β2_adrenergic stimulation. Adenovirus driven expression of human cTnI_wt in adult rat myocytes, monitored by the expression of GFP and specific monoclonal antibodies, does not alter the parameters listed above. As expected, the extent and rate of shortening as well as the relaxation rate of infected and noninfected cardiomyocytes are enhanced in response to isoproterenol (10–5 M) alone and additionally ICI 118,551 (50 nM). Similar effects, but less pronounced, are obtained upon incubation with isoproterenol and CGP_20712A (300 nM).
