Title: Neuronal response abnormalities in the inferior colliculus of the genetically epilepsy-prone rat
Abstract: The genetically epilepsy-prone (GEP) rat is susceptible to seizure induction by acoustic stimuli. The inferior colliculus (IC) has been implicated as being critically important in audiogenic seizure susceptibility based on lesion, electrical stimulation, and focal implantation experiments. The current study determined that GEP rats were most susceptible to seizure induction by pure tone bursts at 100 dB at a frequency of 12 kHz. IC neurons in the GEP rat exhibited a significantly elevated incidence of a particular response pattern at 12 kHz and at characteristic frequency. This pattern consisted of a peak at the beginning and end of the stimulus (onset-offset response). This response pattern only occurred with high intensity stimuli approximating those which induce seizures and may represent an afterdischarge phenomenon. The response threshold was significantly elevated and tuning characteristics were also significantly altered in IC neurons of GEP rats as compared to normal IC neurons. The latter two findings may be related to the deficit of hearing which is reported in the GEP rat. The increased incidence of onset-offset responses may be due to a decreased efficacy of inhibition in the GEP rat neurons as compared to normal rat neurons. Des expériences de 1ésion, de stimulation électrique et d'implantation ont donné une importance critique au colliculus inférieur (CI) dans cette tendance aux crises induites par stimulus auditif. Le présent travail démontre que les rats PGE sont plus particulièrement enclins à présenter une crise si le stimulus est un son pur, donné en bouffées de 100 dB à une fréquence de 12 kHz. Chez le rat PGE, les cellules du CI ont présenté une modalité particulière de réponse à 12 kHz et à la fréquence caractéristique, ceci darts un nombre significativement élevé de cas. Ce pattern était constitué d'un pic au début et à la fin du stimulus (réponse d'installation et de coupure). II n'est apparu qu'avec des stimulus de forte intensité se rapprochant de ceux qui induisent des crises épileptiques et il pourrait refléter un phénoméne de post-décharge. Comparés aux neurones du CI du rat normal, ceux du rat PGE présentaient un seuil de réponse significativement plus élevé, et des caractéristiques d'accord significativement perturbées. Ces deux derniers résultats pourraient étre liés au déficit auditif qui a été décrit chez le rat PGE. L'augmentation de la fréquence d'apparition des réponses d'installation et de coupure pourrait étre due à une diminution de l'efficacité de l'inhibition sur les neurones du rat PGE.
