Abstract: This work involved firstly optimizing the protoplast yields for F. culmorum 159026, then setting up a system for DNA-mediated transformation. Higher protoplast yields and more rapid regeneration were obtained when the organic stabilizers sucrose and sorbitol were used rather than NH4Cl.Successful transformation was achieved using the plasmids pAN7-1 and pHRC containing the E. coli hygromycin B resistance gene (hygromycin B phosphotransferase, hpt) fused to Aspergillus nidulans expression signals.
