Abstract: This chapter describes the history and development of the real-time polymerase chain reaction (PCR). It describes the types of real-time PCR chemistries and analyses the advantages of real-time PCR compared with other techniques. The chapter goes on to look at relevant considerations when designing a real-time PCR assay – such as contamination, controls and reverse transcription real-time PCR. Instrument platforms are covered, including portable real-time PCR detection, with examples of available systems.
