Title: The rate-determining step in the oxidative decarboxylation of isocitric acid
Abstract: The oxidative decarboxylation of isocitric acid by NADP dependent isocitrate dehydrogenase is accompanied by a β-carboxyl carbon isotope effect k12k13 of 0.996 at 25° pH 7.5. This indicates that decarboxylation is not the rate-determining step in this reaction. Some step prior to decarboxylation, probably dehydrogenation, must be at least tenfold slower than decarboxylation.
