Title: Ontogenetic changes in ribulose-1,5-bisphosphate carboxylase-oxygenase activity in soybean leaves
Abstract: Ribulose-1,5-bisphosphate (RuBP) carboxylase and oxygenase (EC 4.1.1.39) activities were measured during ontogenesis of soybean [Glycine max (L.) Merr. cv. ‘Amsoy 71’] leaves. Separate carboxylase and oxygenase assays indicated that the carboxylase activity peaked, whereas the oxygenase activity varied very little over the life of a leaf. The carboxylase to oxygenase ratio was slightly upwardly linear with time. However, simultaneous assays, in which carboxylase activity was estimated radioisotopically while oxygenase activity was estimated polarographically, revealed that the carboxylase to oxygenase ratio increased rapidly during leaf expansion, then remained constant for most of the leaf's functional lifetime before declining as the leaf senesced. The change in the carboxylase to oxygenase ratio seemed to be caused by a greater fluctuation in the carboxylase activity rather than in the oxygenase activity.
