Title: Theoretical effects of binding on transport lag times in the case of multilayered membranes
Abstract: The theoretical solution of the lag time in a general, multilayered membrane (including both binding and partitioning effects) is derived. Comparison of this lag time formula is made to standard lag time formulas derived when only partitioning is taken into account. The special case of the corneal membrane and a single membrane with binding are discussed. It was found that the ratio of low concentration lag times to high concentration lag times is directly proportional to the binding constant of the drug to the membrane. Therefore, the measurement of concentrationdependent lag times can be used to determine binding properties of a membrane with respect to a given drug.
