Title: Modifications in gradient elution of amino acids
Abstract: Analyses of amino acids have been performed mainly by ion-exchange chromatography and special instruments—amino acid analyzers—have been constructed for this purpose, which allows automated analytical runs. The quantitation of amino acids in these instruments is provided by post-column reaction photometric detectors, where ninhydrin as the reagent is added continuously to the eluate from the analytical column and the red products of the reaction of ninhydrin with amino acids are monitored at 570 nm. Columns packed with sulfonated polystyrene divinylbenzene cation-exchange resins and elution with citrate buffers is generally employed. Other liquid chromatographic techniques, in particular reversed-phase chromatography of free or derivatized amino acids and ion-pair chromatography, have been reported to provide rapid separations with excellent efficiency and resolution of the individual amino acids, and these techniques are likely to compete successfully even with ion-exchange chromatography on microparticulate resins or on efficient pellicular or microparticulate chemically bonded exchangers on a silica gel support.
