Title: Combination chemotherapy of malignant glioma. Effect of postoperative treatment with CCNU, vincristine, amethopterine and procarbazine
Abstract: A combination chemotherapy consisting of CCNU, vincristine, amethopterine and procarbazine (COMP-therapy) was studied in the postoperative treatment of malignant gliomas. Fifteen-day COMP-therapy series given 10–20 days after surgery and repeated at regular intervals increased the median recurrence-free interval to 8.9 months and the median survival time to 10.7 months. Both prolongations were significant when compared to the results in a control group not receiving postoperative chemotherapy (median recurrence-free interval: 2.6 months, median survival time: 6.2 months), in a group receiving postoperative CCNU-treatment (3.5 and 6.5 months, respectively), and to a group of patients treated with postoperative polychemotherapy (4.7 and 8.0 months, respectively). The only problematic side effect of COMP-therapy was neuropathy resulting from toxic vincristine-dosage. While nitrosoureas alone were not able to affect the length and the quality of survival, their combination with cell-cycle specific and other cell-cycle non-specific drugs improved the results in the therapy of primary malignant brain tumors.
