Title: 301 The -360T/C Polymorphism in the TNFSF15 Gene Influences the Transcriptional Activity in Activated T Lymphocytes
Abstract: independent analysis of 1850 CD cases and 1817 controls.We replicated CD susceptibility loci in and around 5p13. 1, 5q31.1, 3p21.31,5q33.3 (upstream of IL12B), 10q21, and the ATG16L1, IRGM and NKX2 3 genes, and confirmed recently described associations at 1q24. 3, 6p25.2, and 10p11.21(CCNY).By examining an additional 1103 cases with UC, we could identify the recently suggested CD loci 3p21.31,NKX2-3, CCNY as being associated with UC, too.Whilst the associations of 15q13.1 (HERC2), 17q21.2(STAT3), and 18p11.21(PTPN2) failed to be replicated for CD, they were found to represent novel susceptibility loci for UC.Our findings confirm CD associations from previous GWAS to an unprecedented degree, yield novel associations for UC, and present a comprehensive IBD gene map with multiple variants of modest effect.The overlap of disease gene loci for CD and UC corresponds to similarities observed in terms of the phenotype and of therapeutic responses.
