Title: Scanning Tunnelling Microscopy Investigation of Liposome-DNA-Ca2+ Complexes
Abstract: Liposomes are efficient and reliable systems for gene delivery into target cells. Formation of complexes of liposomes with DNA in the presence of calcium ions was investigated using scanning tunnelling microscopy (STM). Small unilamellar liposomes (SUV) containing egg-phosphatidylcholine, dicetylphosphate (DCP) and cholesterol (CHOL) were prepared. In the presence of DNA extracted from wheat seedlings (Triticum aestivum L. Gerek) and 50 mM Ca2+, the interaction between SUVs was morphologically assessed by STM. The STM images revealed a significant increase in SUV size due to the interaction with DNA and Ca2+, which is an indication of fusion/aggregation of vesicles.
