Title: Specific inhibition of human granulocyte elastase with peptide aldehydes
Abstract: The kinetic features of human granulocyte elastase, chymotrypsin, bovine pancreatic chymotrypsin, porcine pancreatic elastase and elastomucoproteinase were compared. Amino acyi ester substrates were assayed and K m and k cat values were defined. Aldehyde analogues of the P ‐niitroanilide substrates designed for granulocyte elastase as optimal for K m appeared to be potent inhibitors. Suc‐D‐Phe‐Pro‐valinal ( K i = 40μM) was found to inhibit granulocyte elastase competitively and specifically when measured with synthetic substrates, and the K i was 3 μM with the natural protein substrate, elastin.
