Title: Estudio de la TEM con características de célula madre ( o CMC) en pacientes con cáncer de púlmon
Abstract: La Transicion Epitelio-Mesenquima (TEM) juega un papel importante en la migracion celular en procesos fisiologicos normales, pero tambien es fundamental en la progresion del cancer. Ademas, recientemente se ha asociado que las celulas que experimentan una TEM adquieren propiedades de celula madre del cancer (CMC). En este trabajo estudiaremos la expresion de marcadores relacionados con la TEM como E-cadherina y Vimentina, y marcadores relacionados con los dos procesos como Notch1 y Snail2 que son activadores de la TEM y dotan a las celulas con caracteristicas de CMC en pacientes con cancer de pulmon. Utilizamos la tecnica de RT-PCR para ver la expresion del gen Snail2 en sangre periferica de 169 pacientes con cancer de pulmon (CP). Para ver la expresion proteica de E-cadherina, Vimentina, Notch1 y Snail2 en el tejido tumoral de 87 pacientes con CP, utilizamos la tecnica de Inmunohistoquimica. Los programas estadisticos Minitab y SPSS para establecer las distintas correlaciones. Como resultado hemos obtenido que el gen Snail2 se ha expresado en un mayor porcentaje de pacientes con CP en estadios IIIB. En la expresion proteica del tejido tumoral hemos obtenido correlacion significativa de Notch1 con Vimentina y con Snail2. Asi mismo, hemos encontrado un cluster con caracteristicas de CMC e indicios de haber iniciado una TEM principalmente en pacientes con CP celula no pequena y estadios localizados, y con una supervivencia 2,3 veces menor respecto al cluster que no tiene caracteristicas de CMC ni TEM. El aumento de posibles celulas circulantes en mas importante en fases iniciales de la enfermedad. Notch1 dota a las celulas con un fenotipo mesenquimal necesario para iniciar una TEM e impartir propiedades de CMC. En el analisis multivariable del tiempo de supervivencia fueron factores pronosticos independientes de supervivencia el estadio IV de pacientes con CP y el cluster de posibles celulas madre.
