Title: A Tris-Buffered Egg Yolk Extender for Boar Semen
Abstract: Within the range of Tris molarities (0.15, 0.20 and 0.25) and pH levels (6.50, 6.75 and 7.00) investigated, boar spermatozoa were found to tolerate a fairly wide range of molarities and pH's. A phenomenon of sperm tail vibratory movement was observed when boar sperm were stored for over 2 days in extenders of pH 6.75 and 7.00. Such abnormal tail movements were not encountered in extenders of pH 6.50. The addition of catalase was found to eliminate this abnormal tail movement and to improve motility markedly after more than 1 day of storage at 15 C. Glucose, glycine and potassium chloride or their various combinations did not significantly improve sperm survival. The recommended extender consisted of a 0.20 M Tris buffer, containing 3.028 g Tris adjusted to a pH of 7.00 by 1.516 g citric acid monohydrate/100 ml, combined with 20% egg yolk by volume and with 15,000 units of catalase /100 ml and antibiotics added. This extender was generally easy to prepare and could be stored frozen for as long as 8 weeks before use.
