Abstract: This chapter discusses the fluorometric determination of pyridoxal (PAL) and pyridoxal 5′-phosphate in biological materials by the reaction with cyanide. PAL is oxidized to 4-pyridoxolactone, which is a highly fluorescent compound, in the presence of oxygen in slightly alkaline solution by the catalytic action of cyanide. Pyridoxal 5′-phosphate (PAL-P) is hydrolyzed to PAL by acid phosphatase and determined as PAL. The amount of PAL-P is calculated by subtracting the amount of PAL from the value of the sample for PAL-P (hydrolyzed sample) and converting to PAL-P. In the case of the PAL-P determination, it is desirable to correct the determined values by the standard addition extrapolation method.
