Title: Efek ekstrak etanol herba meniran (Phyllanthus niruri L) terhadap organ hati tikus setelah pemberian Karbon Tetraklorida (CCl4)
Abstract: Latar Belakang : Tumbuhan meniran telah dipakai secara empirik oleh masyarakat sebagai hepatoprotektor. Ekstrak P. niruri L. telah dibuktikan efeknya dapat meningkatkan respon imun seluler dan humoral secara in vivo namun demikian pengaruhnya sebagai pemacu regenerasi sel hati belum pernah diteliti. Tujuan : Untuk mengkaji pengaruh pemberian ekstrak etanol herba meniran (P.niruri L.) terhadap kadar enzim hati (SGOT dan SGPT) dan terhadap regenerasi sel hati tikus yang diberi pra perlakuan karbon tetraklorida. Metoda : Hewan coba yang dipergunakan adalah tikus jantan galur Sprague- Dawley dibagi dalam 4 kelompok yaitu: K1 (tanpa perlakuan), K2 (diberi CCl4 2ml/KgBB/hari), kelompok 3 terdiri dari 3 grup yaitu: K3.1 (diberi ekstrak meniran 6 mg/KgBB/hari), K3.2 (diberi ekstrak meniran 20 mg/KgBB/hari), K3.3 (diberi ekstrak meniran 60 mg/KgBB/hari), kelompok 4 terdiri dari 3 grup yaitu: K4.1 (diberi CCl4 dan ekstrak meniran 6 mg/KgBB/hari), K4.2 (diberi CCl4 dan ekstrak meniran 20 mg/KgBB/hari), K4.3 (diberi CCl4 dan ekstrak meniran 60 mg/KgBB/hari). Setiap kelompok dan grup terdiri dari 5 ekor. Ekstrak etanol herba meniran diuji dengan cara diberikan kepada hewan coba peroral selama 30 hari yang sebelumnya diinduksi dengan CCl4 2 ml/KgBB selama 3 hari berturutturut. Sebelum dan setelah perlakuan darah tikus diambil untuk diperiksa kadar SGOT dan SGPT, setelah itu organ hati diambil untuk diproses pengecatan dengan hematoksilin eosin dan secara imunohistokimiawi dengan antibodi terhadap Ki-67. Dilakukan pengamatan terhadap kerusakan sel hati dan proliferasi sel hati yang terwarnai positif Ki-67 secara single blinded. Hasil yang diperoleh dianalisis secara statistik dengan Anova satu jalur. Hasil : Kadar rerata enzim SGOT dan SGPT sebelum perlakuan adalah SGOT 238,32 + 61,16 UI/l dan SGPT 72,22 + 20,13 UI/l, setelah perlakuan kadar rerata enzim SGOT dan SGPT adalah SGOT 109,99 + 82,36 UI/l dan SGPT 77,80 + 46,45 UI/l. Kerusakan sel hati akibat CCl4 2ml/KgBB/hari selama 3 hari pada K2 sebesar 63 + 7,8%, sedangkan rerata persentase kerusakan sel hati pada kelompok K1, K3.1, K3.2, K3.3, K4.1, K4.2 dan K4.3 sebesar 6,77 + 2,70%. Nilai rerata proliferasi sel hati K1: 2,20 + 1,30, K2: 4,00 + 1,41, K3.1: 2,40 + 2,41, K3.2: 2,60 + 1,67, K3.3: 2,40 + 1,67, K4.1: 8,60 + 2,41, K4.2: 9,00 + 7,58 dan K4.3: 19,60 + 7,60. Kesimpulan : Ekstrak etanol herba meniran tidak berpengaruh terhadap kadar enzim hati (SGOT dan SGPT) pasca pemberian karbon tetraklorida, namun mampu menginduksi regenerasi sel hati secara bermakna sesuai dengan pemberian dosis yang diberikan.
Background: Meniran plant has empirically been used as hepatoprotector. The effects of extract of P. niruri L. have been documented to induce cellular and humoral immune responses in vivo. However, its effect on the regeneration of liver cells has not been yet investigated. Objective: To investigate the effects of meniran (P. niruri L.) herbal extract on liver enzymes, namely serum glutamic oxaloacetic transaminase (SGOT) and serum glutamic pyruvic transaminase (SGPT), and liver cell regeneration of the rats pre treated with carbon tetrachloride. Methods: Male rats, Spague-Dawley species, were used in this experiment, and were divided into 4 groups, namely K1 (no treatment), K2 (CCl4 2ml/KgBW/day, K3 consisted of 3 sub groups: K3.1 (6 mg/KgBW/day meniran herbal extract), K3.2 (20 mg/KgBW/day meniran herbal extract, K3.3 (60 mg/KgBW/day meniran herbal extract) and K4 which also consist of 3 sub groups: K4.1 (given CCl4 and 6 mg/KgBW/day meniran herbal extract), K4.2 (given CCl4 and 20 mg/KgBW/day meniran herbal extract), and K4.3 (given CCl4 and 60 mg/KgBW/day meniran herbal extract). Each group and sub groups consist of 5 rats. The meniran herbal ethanolic extract was given orally for 30 days into the rats that had been previously induced with CC14 for 3 days consecutively. Before and after the treatment, the blood of the rats was collected for the SGOT and SGPT assay, the liver organ was prepared for histology and immunohistochemistry using antibody against Ki-67. The result was analyzed statistically using one way Anova. Results: Before treatment, the mean of SGOT enzyme was 238,32 + 61,16 UI/l and the mean of SGPT enzyme was 72,22 + 20,13 UI/l. The mean of SGOT and SGPT enzymes after treatment was 109,99 + 82,36 UI/l and 77,80 + 46,45 UI/l, respectively. The damage of liver rats cells caused by 2 ml/KgBW/day CCl4 for 3 days in K2 was about 63 + 7,8%, while the mean percentage of damage liver rats cells in K1, K3.1, K3.2, K3.3, K4.1, K4.2 and K4.3 were about 6,77 + 2,70%. The mean of proliferation of liver rats cells were K1: 2,20 + 1,30, K2: 4,00 + 1,41, K3.1: 2,40 + 2,41, K3.2: 2,60 + 1,67, K3.3: 2,40 + 1,67, K4.1: 8,60 + 2,41, K4.2: 9,00 + 7,58 and K4.3: 19,60 + 7,60. Conclusion: Meniran herbal ethanolic extract (P. niruri L.) does not affect on liver enzyme levels, but it induces the regeneration of liver cells in dosedependent fashion.
