Title: Ecuaciones cinéticas, momentos estadísticos y tiempos de residencia en los sistemas lineales de compartimentos: aplicación a algunos sistemas enzimáticos
Abstract: El comportamiento cinetico de cualquier sistema lineal de compartimentos (SLC) con inputs cero, cerrado o abierto, con o sin traps viene determinado por el conjunto de ecuaciones diferenciales lineales, de primer orden y coeficientes constantes que vienen dados por las constantes de transferencia fraccional, que describen la variacion de la concentracion de la cantidad de materia. Los Sistemas Enzimaticos en los que la interconexion entre las diferentes formas enzimaticas involucradas en el correspondiente mecanismo de reaccion sean de primer o pseudoprimer orden, pueden ser considerados como casos particulares de SLCs. La resolucion de este sistema de ecuaciones diferenciales, generalmente por el metodo de la transformada de Laplace, para la obtencion de la expresion analitica de la concentracion suele ser una tarea ardua y dificil, y sometido siempre a la comision de posibles errores humanos, por lo que se suele recurrir a integraciones numericas para la resolucion de estos sistemas, sobre todo cuando aumenta el numero de compartimentos y por tanto, el numero de ecuaciones diferenciales.
En esta Tesis Doctoral se presenta una ecuacion general que determina la cantidad de materia en cada compartimento del sistema sin ninguna restriccion ni con respecto a las propiedades de la matriz del correspondiente sistema de ecuaciones diferenciales ni a las multiplicidades de sus valores propios. Su aplicacion puede ser a cualquier sistema quimico, biologico, farmacologico, fisico o cualquier otro sistema que pueda ser modelizado como un SLC, independientemente de su complejidad asi como de su estructura.
Esta ecuacion general puede ser simplificada si se consideran 3 casos comunes en los SLCs y se concretara su aplicacion a tres ejemplos de sistemas enzimaticos de interes fisiologico.
Los momentos estadisticos suelen ser muy utiles para determinar estas constantes de transferencia fraccional. Sin embargo, su uso esta hoy en dia limitado a los momentos de orden cero o de orden uno aplicados a sistemas de compartimentos muy simples En esta Tesis Doctoral, se obtienen las expresiones generales simbolicas, como funcion explicita de las constantes de transferencia fraccional, y para SLCs de cualquier complejidad, y se obtienen a partir de los momentos estadisticos, expresiones unicas para el AUC (Area bajo la curva de niveles plasmaticos), el AUMC (Area bajo la curva del primer momento) y el Tiempo de Residencia Medio (MRT) en funcion de las constante de transferencia.
Ademas se presenta en esta Tesis Doctoral un software unico en el mundo llamado bioCelTer que calcula el MRT. Este software es aplicable a cualquier sistema de compartimentos y nos proporciona tanto las expresiones simbolicas como el valor numerico del MRT en un organo del sistema o en la totalidad en su conjunto.
Finalmente se aplica bioCelTer a sistemas enzimaticos, ya que a partir de los Tiempos de Residencia Medios se puede obtener el tiempo de actividad enzimatica, que es el tiempo durante el cual un sistema tiene actividad enzimatica, parametro que no ha sido calculado hasta ahora, debido a la dificultad de los calculos a realizar.
