Abstract: Introdução: A leishmaniose tegumentar americana (LTA) no Brasil apresenta-se em fase de expansão geográfica nos últimos anos.Com o aumento do número de casos humanos, testes sorológicos vêm sendo aprimorados no intuito de auxiliar o diagnóstico das leishmanioses além de serem importantes ferramentas em inquéritos epidemiológicos.Objetivo: Avaliar a especificidade e sensibilidade do teste ELISA utilizando antígenos de diferentes espécies de Leishmania.Material e Métodos: Soros de pacientes saudáveis (49), LTA (60), leishmaniose visceral (30), Malária (32), Doença de Chagas (49) e Tuberculose (13) foram analisados utilizando-se antígenos solúveis de diferentes espécies de Leishmania.As espécies utilizadas foram Leishmania (Leishmania) amazonensis -IFLA/BR/67/PH8 e Leishmania (Viannia) braziliensis -isolado da região de Uberlândia MG.Os antígenos de promastigotas foram preparados em diferentes tampões quanto a constituição e pH.Resultados: Os antígenos de L. amazonensis e L. braziliensis, preparados com tampão fosfato pH 8,0 apresentaram sensibilidade de 85% e 93% e especificidade de 97% e 88%, respectivamente.Em tampão Tris pH 7,4, apresentaram sensibilidade de 42% e 92% e especificidade de 90% e 93%, respectivamente.Conclusão: Os maiores índices de sensibilidade observados no teste ELISA com antígeno solúvel de Leishmania braziliensis, sugerem que o reconhecimento sorológico espécie-específico de epítopos presentes na cepa utilizada, visto que a grande maioria das amostras de soro de LTA eram de pacientes infectados por Leishmania (Viannia) braziliensis.Portanto, a utilização de preparados antigênicos utilizando isolados de Leishmania responsáveis pela infecção em determinadas regiões, podem aumentar os níveis de anticorpos específicos para LTA.
