Title: Significance of expression of bcl-2 mRNA and bax mRNA in arsenic trioxide-induced apoptosis of hepatocellular carcinoma cells
Abstract: ObjectiveTo explore of significance of expression of bcl-2 mRNA and bax mRNA in arsenic trioxide-induced apoptosis of hepatocellular carcinoma (HCC) cells. MethodsAfter human HCC cell line bel-7402 was treated with arsenic trioxide at various concentrations, the apoptosis was detected with transmission electron microscopy, flow cytometry and DNA laddering. Meanwhile, the expression of bcl-2 mRNA and bax mRNA was determined with semiquantitative reverse transcription polymerase chain reaction (RT-PCR). ResultsCell apoptosis was not detected in 0.5 μmol/L As_2O_3 group but in 8 μmol/L As_2O_3 group. The expression of bcl-2 mRNA were not seen in the control group and drug-treated group. Enhancement of expression of bax mRNA along with increase of As_2O_3 concentration. ConclusionsProliferation is inhibited via inducing apoptosis by As_2O_3 in bel-7402 cell line. Enhancement of bax mRNA expression but not that of bcl-2 mRNA expression is related to this process.
