Title: Isolation and sequence analysis of B-class MADS-box gene in sweet cherry
Abstract: To study the role of MADS-box genes in floral development of sweet cherry,MADS-box gene was isolated from Prunus avium L.Lapins with RT-PCR and RACE approaches.Primers were designed according to the conservative region sequences of various plant MADS-box family genes,and semi-quantitative PCR was used to analyze the expression of gene in different organs and tissues.The full length of cDNA is 962 bp,named PaMADS2(the accession numbers in GenBank: HQ229606),with an open reading frame(ORF) of 633 bp and encoding 210 amino acid residues.Sequence comparison and phylogenetic analysis indicated that PaMADS2 closely resembles the PI clade of class B.Tissue specific analysis showed that PaMADS2 expressed in petal and stamen.The results suggested that PaMADS2 belongs to the B-class MADS-box gene family.
