Title: Opimization of conditions for extracting astaxanthin from Rhodozyma
Abstract: It was studied for extracting intracellular astaxanthin from Rhodozyma in this experiment.Firstly,the effects of DMSO concentration,cell disruption temperature,cell disruption time on cell disruption were determined,respectively;Sencondly,extracting process opimization was carried out based on extraction solvent,extraction temperature,extraction time and pH condition.The conditions for extracting astaxanthin from Rhodozyma optimized by orthogonal experiment.The results showed that the optimum disrupting conditions were: DMSO concentration 100%,cell disruption temperature 40 ℃,cell disruption time 30 min.It indicated that the optimum extracting conditions of astaxanthin were: extraction solvent DMSO:absolute alcohol(3:2,v:v),extraction temperature 40 ℃,extraction time 60 min and pH 3.Conclusions: the method was simple and good that astaxanthin from Rhodozyma was extracted by DMSO with absolute alcohol.
