Title: Effect of Anti-TNF-α Monoclonal Antibody on Rat Ulcerative Colitis
Abstract: Objective To investigate the effect of anti-TNF-α monoclonal antibody(mab)on ulcerative colitis(UC) in rats.Methods Thirty male Sprague-Dawley(SD) rats were randomly divided into three groups:normal control group,UC control group and treatment group,10 in each.A solution containing ethanol and 2,4,6-trinitrobenzenesulfonic acid(TNBS) was instilled into the distal colon in the rats,in a dose of 100 mg/kg,to establish acute models of UC rats.The treatment group were treated with anti-TNF-α mab,UC and normal control groups with caudal intravenous injection of 0 9% sodium chloride solution.All rats were sacrificed simultaneously after 6 days of treatment.We observed their colonic mass,colon myeloperoxidase(MPO) activity,serum levels of TNF-α and expression of TNF-α mRNA,to evaluate degree of inflammation and injury.Results As compared with normal control group,ratio of colonic mass/body mass(mc/mb) and activity of colonic MPO were markedly increased in UC control group(P0.01),and a significant increase in both serum TNF-α level and colonic TNF-α mRNA expression was noted(P0.01).After treatment,a sharp reduction in mc/mb and MPO activity was demonstrated in treatment group as compared with UC control group(P0.01).Serum TNF-α level was decreased,and colonic TNF-α mRNA expression inhibited by anti-TNF-α mab(P0.01).Furthermore,these parameters were highly correlated with each other(P0.01).Conclusion The increase in serum TNF-α level and colonic TNF-α mRNA expression is in sympathy with the disease severity.TNF-α is closely involved in the immune abnormalities and inflammatory responses in UC.High level of TNF-α and its corresponding mRNA expression seem to be implicated in the pathogenesis of UC.Anti-TNF-α mab has therapeutic effect on UC by down-regulating serum TNF-α and colonic TNF-α mRNA expression.
