Title: Pharmacokinetics of extended-release tablets of loxoprofen in Beagle dogs
Abstract: Aim:To establish an LC-UV method for the bioavailability and pharmacokinetics study of loxoprofen in Beagle dogs.Methods:After adding ketoprofen(internal standard) and 1 mol/L HCL 100 μL,1 mL of plasma were extracted with 5 mL of ethyl acetate and determined by LC-UV.A random two-way crossover study in 8 dogs was adopted.The parameters of pharmacokinetic were calculated by BAPP 2.2 program.Results:The results indicated that the t_(max) and t_(1/2) of extended-release tablets(ER) were longer than those of immediate-release tablets(IR),and the c_(max) of ER was lower than that of IR: The bioavailability of ER relative to IR was(95.2±19.0)%.Conclusion:The method was proved to be simple,sensitive and rapid.The two formulations had the same absorption extent.Comparing with IR,the ER exhibited good extended release tendency.
