Title: The Effect of Sheng-xue-ling on the Protein Expression of Steroid-resistant ITP Patients
Abstract: Objective: To observe the effect of Sheng-xue-ling on protein expression in steroid-resistant immune thrombocytopenia(ITP) as well as its mechanism of action.Methods: With such proteomics technology as two-dimensional electrophoresis and mass spectrometry it was established who were the profiles of serum total protein two-dimensional gel electrophoresis(2-DE) of the ITP patients and of the steroid-resistant ITP patients.Then the difference of protein loci was analyzed between each group.And the effect of Sheng-xue-ling was observed on protein expression in steroid-resistant immune thrombocytopenia(ITP) patients.Results: Among steroid-resistant ITP patients the protein expression of cellobiose hydrolytic enzymes was high,and that of exocellobiohydrolase was low.There was significant difference between the ITP patients and the steroid-resistant on them(P0.05).After treatment with sheng-xue-ling,the protein expressions were improved,and a significant difference showed up compared with before treatment(P0.01).Conclusion: The high protein expression of cellobiose hydrolytic enzymes and the low protein expression of exocellobiohydrolase could be the important action mechanism of hormonal resistance in ITP patients,which could be also the important cause of ineffective hormone therapy.Sheng-xue-ling can improve the clinical efficacy through regulating the protein expression and the reversal of hormone resistance. Shengxue Ling by regulating the protein expression level and,improve the clinical efficacy.
