Title: Studies on Wheat Genetic Transformation Mediated Via Agrobacterium-tumefaciens in Which the Mature Embryos to be Explants
Abstract: In this study,the genetic transformation procedure of wheat mediated via Agrobacterium-tumefaciens in which the mature embryos to be the explants was established.Under selection pressure condition,the generation rate of resistant calli to the transformed total mature embryos was 57.50%,with a differentiation rate of resistant buds to the transformed total mature embryos being 8.58%,and a plant generation rate of generated plants to the transformed total mature embryos being 0.83%.In the tested three generated plants,PCR amplifications for the reporter gene Gus were all positive.In the meantime,the Gus activities in the plants were all significantly higher than the control(non-transformed plants).These results demonstrated that the tested plants generated from the transformed mature embryos were transgenic.Therefore,the genetic transformation approach established in this study would provide a powerful method for establishment of transgenic wheat lines in the future.
