Title: Activation condition of NF-κB in leukemia cells and its effect on cellular apoptosis
Abstract: Objective To investigate the activation condition of NF-κB in leukemia cells and its effect on cellular apoptosis.Methods Flow cytometry was used to detect the apoptosis of leukemia cells and peripheral blood mononuclear cell(PBMCs).The relative activities of NF-κB in the cells were assessed by luciferase reporter assays.After treated by PDTC(the inhibitor of NF-κB) for 12 h,the activation condition of NF-κB and apoptosis in leukemia cells were analyzed.In addition,the mRNA and protein expression of Bax and Bcl-2 were analyzed by qPCR and Western blot,respectively.Results Compared with PBMCs,cellular apoptosis in leukemia cells were significantly decreased(P0.05).The results of luciferase reporter showed that the relative activities of NF-κB were tly higher in leukemia cells than PBMCs(P0.05).Upon PDTC treatment for 12 h,the relative activities of NF-κB in leukemia cells were significantly decreased,while cellular apoptosis were significantly increased(P0.05).Additionally,the mRNA and protein expression of Bax in leukemia cells were significantly increased(P0.05),while the mRNA and protein expression of Bcl-2 were remarkably decreased(P0.05).Conclusion Constitutively activated NF-κB exists in the leukemia cells,and it's constitutively activation induces cellular apoptosis resistance.
