Title: Effect of microRNA-15b on the apoptosis of drug-resistant cells of lung cancer via down-regulating Bcl-2
Abstract: Objective To investigate the effect of miRNA-15b on the apoptosis of drug-resistant cells of lung cancer A549/DDP.Methods MiR-15b in vitro was transiently transferred into A549/DDP by FuGENE-HD.The expression of MiR-15b in A549/DDP cells was detected by real-time fluorescence quantitative PCR,and the apoptosis of A549/DDP cells was analyzed by flow cytometry.The expression of Bcl-2 protein was detected by Western blot.Results The expression of miR-15b in miR-15b-transfected A549/DDP cells increased significantly( P 0.05).The apoptosis of transfected cells was 32.4% ± 5.1%,5.73% ± 1.2% and 6.24% ± 2.4% respectively in the miR-15b group,the Mock group and the miR-15b-Cont group,which showed significant differences( P 0.05).The expression of Bcl-2 increased more obviously in the miR-15b group than in the control group( P 0.05).Conclusion The miR-15b may induce the apoptosis of A549/DDP cells by down-regulating the expression of Bcl-2,which provides a new target for the treatment of lung cancer.
