Title: Study on the Relationship between P13K/Akt Signal Transduction Pathway and Cisplatin Resistance in Ovarian Carcinoma
Abstract: Objective: To investigate the relationship between cisplatin (DDP) resistance of ovarian carcinoma and PI3K/Akt signal transduction pathway. Methods: SKOV3 and SKOV3/DDP cells were divided into 6 groups depending on the concentration of LY294002, which were 5 μmol/L,10 μmol/L,20 μmol/L,40 μmol/L,DDP and control group. MTT assay was used to analyze the effect of 40 μmol/L LY294002 on the sensitivity of SKOV3 and SKOV3/DDP. Western blot was used to detect the expressions of PI3K, Akt and β-actin in SKOV3 and SKOV3/DDP cells of each group. Results: After interference by 40 μmol/L of LY294002, the sensitivity to DDP of SKOV3 and SKOV3/DDP cells increased 67.36% and 76.56%. The expressions of PI3K and Akt in SKOV3 and SKOV3/DDP cells were reduced by LY294002. The correspondence densities of PI3K and Akt were lower in SKOV3 cells than those in SKOV3/DDP cells (t = 26.005 and t = 23.477,P 0.05).Conclusion: The abnormal PI3K/Akt signaling pathway is related to cisplatin resistance in ovarian carcinoma. The chemosensitivity of ovarian cancer can be enhanced by inhibiting PI3K/Akt signaling pathway.
